The elastin fiber network in the dermis enables the skin to stretch and recoil. Unlike other extracellular matrix (ECM) components such as collagen and hyaluronic acid, which are continually synthesized and assembled through life, elastic fibers are primarily synthesized during late gestation and early childhood, have a half-life of about 70 years, and are therefore considered to persist through an individual's lifespan. However, due to common aging processes, accelerated by environmental factors, elastic fibers are enzymatically degraded through elastases and cannot be repaired or replaced properly. Severe enzymatic degradation of elastic fibers can lead to a wide range of congenital and acquired skin elastic fiber disorders, such as mid-dermal elastolysis and cutis laxa.
